Tadalafil ek phosphodiesterase type 5 (PDE5) inhibitor hai jo mainly erectile dysfunction (ED), benign prostatic hyperplasia (BPH), aur pulmonary arterial hypertension (PAH) ke treatment ke liye use hota hai. Ye medicine blood vessels ko relax karke unmein khoon ke flow ko improve karta hai.

Tadalafil PDE5 enzyme ko inhibit karta hai, jo cyclic guanosine monophosphate (cGMP) ke breakdown ke liye responsible hota hai.
Isse:

  • Erectile function improve hoti hai (ED mein)
  • Urinary symptoms relieve hote hain (BPH mein)
  • Pulmonary vascular resistance kam hoti hai (PAH mein)

Contraindications:

  • Nitrate medications ke saath use (angina ke treatment mein): severe hypotension ka risk hota hai.
  • Severe cardiovascular disorders (e.g., recent heart attack or stroke)
  • Known hypersensitivity ya allergy to Tadalafil or its excipients
  • Severe liver or kidney impairment

Precautions:

  • Cardiovascular risk patients (especially during sexual activity)
  • Alpha-blockers ke saath caution (postural hypotension ka risk)
  • Visual impairment history (non-arteritic anterior ischemic optic neuropathy – NAION)
  • Hearing loss: sudden hearing decrease reported
  • Nitrates (e.g., nitroglycerin): severe drop in blood pressure

  • Alpha-blockers (e.g., tamsulosin): symptomatic hypotension risk

  • CYP3A4 inhibitors (e.g., ketoconazole, ritonavir): Tadalafil levels increase

  • CYP3A4 inducers (e.g., rifampin): Tadalafil efficacy reduce ho sakti hai

  • Alcohol: excessive use se blood pressure drop aur dizziness ho sakti hai

Common Side Effects:

  • Headache

  • Dyspepsia (indigestion)

  • Back pain

  • Muscle aches

  • Nasal congestion

  • Flushing (redness of face)

Less Common / Serious Side Effects:

  • Vision changes (e.g., blurred vision, color distortion)

  • Hearing loss

  • Hypotension

  • Priapism (prolonged erection)

  • Allergic reactions (rash, swelling, difficulty breathing)

  • Pregnancy: Tadalafil ka use pregnant women mein recommended nahi hai, kyunki iska safety profile in populations clearly established nahi hai (Category B – animal studies show no risk, but human data lacking).

  • Breastfeeding: Unknown whether tadalafil is excreted in human milk. Caution advised. Breastfeeding mothers ko use avoid karna chahiye jab tak clear safety data available na ho.

Store this medication at 68°F to 77°F (20°C to 25°C) and away from heat, moisture and light. Keep all medicine out of the reach of children. Throw away any unused medicine after the beyond use date. Do not flush unused medications or pour down a sink or drain.
